Effects of hrb1/SPAC328.05 on mRNA export in fission yeast

Abstract

|
In fission yeast, Schizosaccharomyces pombe, we previously reported the isolation of mutant (SLrsm1), which showed a synthetic lethality in combination with the rsm1 null allele, to identify genes necessary for mRNA export. Here, we demonstrate that the hrb1/SPAC328.05 gene is isolated, which partially suppresses the defects of growth and mRNA export of SLrsm1. This gene encodes an RRM (RNA recognition motif)-containing protein that is homologous to Saccharomyces cerevisiae Gbp2 and Hrb1 involved in mRNA surveillance and nuclear mRNA quality control. The deletion of hrb1/SPAC328.05 did not cause any defects of growth and mRNA export. However, its overexpression exhibited severe growth defect and accumulation of poly(A)+ RNA in the nucleus. Moreover, we observed that the Hrb1-GFP protein is localized predominantly in the nucleus. These observations support the possibility that the S. pombe Hrb1/SPAC328.05 protein is also involved in mRNA export and quality control.
